System Dispersion in (U)HPLC is the reason why peaks can look ‘fatter’. This can effect resolution (separation of peaks) and the sensitivity (lowest limit of detection or quantitation).
You can control the extra-column dispersion and limit its effect. The Intra-column dispersion is dependent on what column you choose and the packing etc (usually outside your control).
What to choose for the best Extra Column Dispersion performance
Did you know about?
Shimadzu Nexflow technologies
The columns used in ultra high performance liquid chromatography (UHPLC) are the cornerstone of high performance, high resolution analysis that achieves excellent chromatographic separation with minimal dispersion in the column. However, the phenomenon of peak band dispersion outside the column often offsets the concentration effect of the component peaks obtained in the column, resulting in the deterioration of peak shape and separation
Nexflow technology consists of two components to provide the best performance:
・Proprietary shape piping
・Low-dispersion injection mode of the autosampler

Advanced User Information
Low dispersion Injection mode
NB/ Full volume injection method used as standard in existing Nexera
A low-dispersion injection mode has been added to the SIL injection method to prevent the dispersion of sample bands. Switch to the low-dispersion injection mode with the checkbox on the LabSolutions screen shown in Figure 4. However, in the low-dispersion injection mode, the injection time is increased because the sample band dispersion is suppressed*1.
*1: The injection time is increased by approximately 10 seconds, but the injection time is maintained at the same level as that of the competitors.
The Result of Effects using Shimadzu Nexflow Technology
